Overview

AZD1043 is a long-acting Peptide YY (PYY) analogue, specifically a PYY3-36 mimetic, developed by AstraZeneca for the treatment of obesity and overweight. PYY is a gut-derived peptide hormone that is naturally released from the gastrointestinal tract postprandially to signal satiety to the brain. AZD1043 selectively targets and activates the Neuropeptide Y receptor type 2 (NPY2R) in the central nervous system, particularly within the hypothalamus and hindbrain, to reduce appetite and caloric intake. By utilizing molecular modifications to extend its half-life, AZD1043 is designed for infrequent administration to provide sustained weight loss efficacy. It is currently in Phase I clinical development, where it is being evaluated for safety, tolerability, and pharmacokinetics via subcutaneous and intravenous administration in individuals living with overweight or obesity.
