Overview

AZD1222 + rAd26-S is a heterologous prime-boost COVID-19 vaccine regimen that combines two distinct adenoviral vector platforms. The regimen consists of one dose of AZD1222 (also known as ChAdOx1 nCoV-19 or Vaxzevria), a non-replicating chimpanzee adenovirus vector vaccine developed by AstraZeneca and the University of Oxford, and one dose of rAd26-S, a recombinant human adenovirus type 26 vector vaccine that serves as the first component of the Sputnik V vaccine developed by the Gamaleya Research Institute. Both vaccines are engineered to deliver the genetic sequence for the SARS-CoV-2 spike glycoprotein to host cells, inducing both humoral and cellular immune responses. This combination was primarily evaluated in Phase I/II clinical trials to determine if mixing different adenoviral vectors could enhance immunogenicity and safety while potentially mitigating the impact of anti-vector immunity.
