Overview

AZD1419 is a synthetic oligonucleotide and a potent toll-like receptor 9 (TLR9) agonist developed by AstraZeneca in collaboration with Dynavax Technologies. It was designed as an inhaled immunostimulant to modulate the immune response in asthma and potentially chronic obstructive pulmonary disease (COPD). The drug acts by stimulating TLR9, leading to activation of the type I interferon pathway and shifting immune responses from Th2-dominant (allergic/inflammatory) toward Th1-dominant profiles. In preclinical models and early clinical studies, AZD1419 induced dose-dependent production of IFN-α and suppressed allergic cytokines such as IL-5. Despite demonstrating target engagement, reduction in type 2 inflammation markers, and safety/tolerability in phase I/II trials, it did not lead to improved asthma control compared to placebo. Development for both asthma and COPD has been discontinued[1][3][7][8].
