Overview

AZD1446 is a highly selective small molecule agonist of the α4β2 subtype of neuronal nicotinic acetylcholine receptors (nAChRs). It was developed as a non-desensitizing nicotinic agonist to target cognitive disorders and neurological diseases by modulating cholinergic neurotransmission in the brain. Preclinical and clinical studies have explored its potential for treating conditions such as attention deficit-hyperactivity disorder (ADHD), Alzheimer's disease (as adjunct to donepezil), and movement disorders like dystonia and levodopa-induced dyskinesia. While AZD1446 demonstrated good safety and tolerability profiles in early-phase trials, it did not show efficacy in ADHD or DYT1 dystonia models but showed some pro-cognitive signals that supported further investigation for Alzheimer's disease[1][5][6][10].
