Overview

AZD2373 (IONIS-AZ4-L) is an investigational ligand-conjugated antisense (LICA) oligonucleotide designed to reduce the production of coagulation Factor XI (FXI). Developed through a collaboration between Ionis Pharmaceuticals and AstraZeneca, the drug targets FXI mRNA in the liver, utilizing Ionis's proprietary GalNAc (N-acetylgalactosamine) conjugation technology to enhance hepatocyte-specific delivery and potency. By inhibiting the hepatic synthesis of FXI, AZD2373 aims to provide a localized anticoagulant effect that prevents thrombosis while minimizing the systemic bleeding risks associated with traditional anticoagulants. The development program primarily focused on patients with end-stage renal disease (ESRD) undergoing hemodialysis, who are at high risk for both thrombotic events and bleeding complications. However, as of 2023, AstraZeneca has removed AZD2373 from its clinical development pipeline, effectively terminating the program.
