Overview

AZD4045 is an investigational allogeneic chimeric antigen receptor T-cell (CAR-T) therapy developed by AstraZeneca for the treatment of relapsed or refractory multiple myeloma (RRMM). It specifically targets the B-cell maturation antigen (BCMA), a protein highly expressed on the surface of malignant plasma cells. As an allogeneic therapy, AZD4045 is derived from healthy donor T-cells rather than the patient's own cells, potentially offering an 'off-the-shelf' treatment option that bypasses the complex and time-consuming manufacturing process associated with autologous CAR-T therapies. The drug is currently being evaluated in a modular Phase I/II clinical trial as a monotherapy and in combination with other agents, including daratumumab (an anti-CD38 monoclonal antibody) and aldesleukin (recombinant interleukin-2), to assess its safety, tolerability, and preliminary efficacy in patients with advanced multiple myeloma.
