AZD4547 + anastrozole or letrozole is a combination therapy investigated for the treatment of estrogen receptor-positive (ER+) metastatic breast cancer that has become resistant to aromatase inhibitors. AZD4547 is a potent and selective small molecule inhibitor of fibroblast growth factor receptor (FGFR) 1, 2, and 3 tyrosine kinases. Anastrozole and letrozole are non-steroidal aromatase inhibitors that reduce estrogen production by inhibiting the aromatase enzyme. The rationale for this combination is that FGFR pathway activation can mediate resistance to endocrine therapy in breast cancer; thus, combining FGFR inhibition with continued estrogen suppression may overcome resistance mechanisms. Clinical trials have shown this combination to be generally safe with manageable side effects and some evidence of anti-tumor activity in heavily pretreated patients[1][3][6].
