Overview

AZD5069 + durvalumab + tremelimumab is a combination therapy involving three investigational and/or approved drugs: AZD5069, a small molecule selective antagonist of the CXC chemokine receptor 2 (CXCR2); durvalumab, a monoclonal antibody and immune checkpoint inhibitor targeting programmed death-ligand 1 (PD-L1); and tremelimumab, a monoclonal antibody checkpoint inhibitor targeting cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4). The combination is being investigated for synergistic anti-tumor effects by inhibiting pro-tumor immune signaling (via CXCR2), blocking immunosuppressive signals (PD-L1), and promoting T-cell-mediated anti-tumor immunity (CTLA-4). It is in clinical studies for advanced solid tumors, hepatocellular carcinoma (HCC), and recurrent/metastatic squamous cell carcinoma of the head and neck. The combination is currently in early-phase clinical trials.
