Overview

AZD5423 is a nonsteroidal, small molecule selective glucocorticoid receptor modulator (SGRM) developed by AstraZeneca for the treatment of respiratory diseases, primarily asthma and chronic obstructive pulmonary disease (COPD). It acts as a glucocorticoid receptor agonist but is structurally distinct from traditional steroidal glucocorticoids. The drug was designed to provide anti-inflammatory effects in the airways with potentially fewer systemic side effects compared to conventional corticosteroids. Clinical studies demonstrated that inhaled AZD5423 could attenuate allergen-induced airway responses and reduce sputum eosinophilia in asthma patients, but it did not show clinically meaningful improvement in lung function for COPD patients at tested doses. Development for both asthma and COPD has been discontinued[1][5][6][7][8][10].
