Overview

AZD5851 is an autologous chimeric antigen receptor T-cell (CAR-T) therapy under development for the treatment of glypican-3 (GPC3) positive advanced or recurrent hepatocellular carcinoma. The therapy consists of a patient's own T lymphocytes genetically modified to express a CAR specific for GPC3, a protein overexpressed in hepatocellular carcinoma but largely absent from healthy tissues. AZD5851 also incorporates a dominant negative transforming growth factor beta receptor II (dnTGFβRII) as an armoring strategy to block TGFβ signaling and protect CAR-T cells from immunosuppressive effects within the tumor microenvironment. The drug is being developed by AstraZeneca and AbelZeta Pharma and is currently in Phase 2 clinical trials for hepatocellular carcinoma[2][3][4][5].
