Overview

AZD6422 is an autologous, armored chimeric antigen receptor T cell (CAR-T) therapy designed to target Claudin 18.2 (CLDN18.2), a tumor-associated antigen highly expressed in certain gastrointestinal cancers such as gastric, pancreatic, and esophageal tumors[1][3][5]. The therapy uses T lymphocytes genetically modified to express a CAR specific for CLDN18.2 and incorporates dominant-negative TGF-β receptor armoring to enhance antitumor activity by counteracting immunosuppressive signals in the tumor microenvironment[3][5][6]. The construct includes CD28 and CD3z costimulatory domains for improved activation and persistence of the engineered T cells[6]. Preclinical studies have shown significant antitumor efficacy and tolerability in patient-derived xenograft models with various levels of CLDN18.2 expression and TGF-β signaling[3][5][6]. AZD6422 is currently being evaluated in Phase 1 clinical trials for advanced or metastatic CLDN18.2-positive gastrointestinal tumors[4].
