Drug intelligence / Profile preview
AZD6621 is an experimental, next-generation tri-specific T-cell engager (TCE) developed by AstraZeneca for the treatment of metastatic castration-resistant prostate cancer (mCRPC). It is designed to target six-transmembrane epithelial antigen of prostate-2 (STEAP2) on prostate cancer cells, while simultaneously engaging CD3 and CD8 on T cells. By incorporating a CD8-guided, affinity-optimized CD3 engagement mechanism, AZD6621 preferentially activates CD8+ cytotoxic killer T cells over CD4+ helper T cells. This selective activation is intended to maximize tumor cell destruction while minimizing systemic inflammatory cytokine release, thereby improving the therapeutic index compared to conventional T-cell engagers. AZD6621 is currently being evaluated in a Phase I/II clinical trial (ACTIVATED-4-PC) as a monotherapy for patients with metastatic prostate cancer.
