AZD7003 (also known as C-CAR031) is an autologous, armored chimeric antigen receptor T-cell (CAR-T) therapy designed to target Glypican-3 (GPC3), a cell-surface oncofetal antigen highly expressed in hepatocellular carcinoma (HCC) and other solid tumors. Developed through a collaboration between AstraZeneca and AbelZeta Pharma, the therapy utilizes AstraZeneca's transforming growth factor-beta receptor II (TGFβRII) dominant negative armoring platform. This modification allows the CAR-T cells to resist the immunosuppressive effects of TGFβ within the solid tumor microenvironment, potentially enhancing persistence and anti-tumor activity. AZD7003 is currently being evaluated in Phase I/II clinical trials for the treatment of advanced GPC3-positive HCC and squamous non-small cell lung cancer.
