AZD7624 is an inhaled small molecule inhibitor of mitogen-activated protein kinase 14 (p38α), developed by AstraZeneca for the treatment of chronic obstructive pulmonary disease (COPD). It selectively inhibits the α and β isoforms of p38 MAPK, with no activity against γ and δ forms. Preclinical studies demonstrated potent inhibition of human MAPK14 and suppression of TNFα release from human peripheral blood mononuclear cells. Inhaled administration leads to rapid absorption and high local lung exposure with minimal systemic effects. Despite promising pharmacodynamic effects in reducing inflammatory markers in early clinical studies, AZD7624 failed to show clinical benefit in COPD exacerbation prevention trials, leading to discontinuation of its development for this indication[1][5][7].
