Overview

AZD7762 + irinotecan is a combination of a **selective checkpoint kinase inhibitor** (AZD7762) and a **topoisomerase I inhibitor** (irinotecan) developed for the treatment of advanced solid tumors. **AZD7762** is a potent, selective ATP-competitive inhibitor of Chk1 (checkpoint kinase 1), which disrupts cell cycle arrest in response to DNA damage, thereby potentiating the cytotoxicity of DNA-damaging agents. Irinotecan inhibits topoisomerase I, preventing DNA re-ligation and causing cell death. Preclinical studies have shown that AZD7762 enhances the effect of irinotecan and its active metabolite SN-38. This combination was investigated in a Phase I open-label, dose-escalation clinical trial in patients with advanced solid malignancies, administered via intravenous infusion. The trial was sponsored and developed by AstraZeneca.
