Overview

AZD8076 is a DNA-encoded monoclonal antibody (DMAb) developed through a collaborative research program involving AstraZeneca, INOVIO, The Wistar Institute, and the University of Pennsylvania. It is designed to provide passive immunity against COVID-19 by delivering synthetic DNA plasmids that encode for a neutralizing antibody derived from AstraZeneca's cilgavimab or tixagevimab (the components of Evusheld). The therapy is administered using INOVIO's CELLECTRA electroporation device, which facilitates the entry of the DNA plasmids into host cells, where the host's cellular machinery then produces and secretes the functional monoclonal antibodies in vivo. This approach aims to offer a durable, scalable, and tolerable alternative to traditional protein-based antibody therapies, potentially providing sustained protection with fewer administrations. AZD8076 was evaluated alongside AZD5396 in a Phase 1 proof-of-concept trial funded by DARPA and other defense agencies.
