Overview

AZD8154 is a novel, selective, inhaled dual inhibitor of the gamma (γ) and delta (δ) isoforms of phosphoinositide 3-kinase (PI3K), developed by AstraZeneca for the treatment of inflammatory respiratory diseases such as asthma. By targeting both PI3Kγ and PI3Kδ, the drug aims to reduce pulmonary inflammation driven by both Th1/Th17 and Th2 immune pathways, potentially restoring corticosteroid sensitivity in patients with severe disease. It is designed for once-daily inhaled administration, featuring low solubility for prolonged lung retention and minimal systemic exposure to mitigate the side effects typically associated with oral PI3K inhibitors.
