Overview

AZD8421 is an orally bioavailable, highly selective small molecule inhibitor of cyclin-dependent kinase 2 (CDK2), developed by AstraZeneca for the treatment of advanced or metastatic solid tumors, including ER-positive/HER2-negative breast cancer and ovarian cancer. CDK2 plays a crucial role in cell cycle progression and is implicated in resistance mechanisms to CDK4/6 inhibitors as well as in cancers with CCNE1 amplification. By inhibiting CDK2, AZD8421 aims to slow or stop the proliferation of cancer cells, particularly those that have become resistant to other therapies. The drug is currently being evaluated in Phase I/II clinical trials both as monotherapy and in combination with other targeted anti-cancer agents such as camizestrant (a SERD) and CDK4/6 inhibitors (abemaciclib, ribociclib, palbociclib)
