Overview

AZD8529 is a potent and selective positive allosteric modulator (PAM) of the metabotropic glutamate receptor 2 (mGluR2). It enhances the effects of glutamate at mGluR2 with high specificity and minimal off-target activity. Preclinical studies demonstrated that AZD8529 reduces psychomotor activity and neural firing in rodent models, and it has shown efficacy in reducing nicotine self-administration and relapse in non-human primate models. The drug was developed by AstraZeneca primarily for central nervous system indications such as schizophrenia and nicotine addiction. In clinical trials for schizophrenia, however, it did not demonstrate significant antipsychotic efficacy compared to placebo[1][3][4][5].
