Overview

AZD9773 (CytoFab) is a first-in-class, ovine (sheep-derived) polyclonal antibody fragment (Fab) product that targets and neutralizes tumor necrosis factor alpha (TNF-alpha), a key pro-inflammatory cytokine implicated in the pathophysiology of sepsis and septic shock. Unlike monoclonal antibodies, AZD9773 consists of multiple Fab fragments derived from sheep immunized with human TNF-alpha, resulting in broad reactivity against this target. The drug was developed for intravenous administration to rapidly reduce circulating TNF-alpha levels in patients with severe sepsis or septic shock. Clinical studies demonstrated that while AZD9773 efficiently decreased plasma TNF-alpha concentrations, this pharmacodynamic effect did not translate into improved clinical outcomes such as ventilator-free days or mortality reduction compared to placebo[2][6][8]. Development has been discontinued for all indications.
