Overview

Azelastine + epinastine is a hypothetical combination of two second-generation antihistamines, each with dual mechanisms of action. Azelastine is primarily a histamine H1-receptor antagonist used intranasally and ophthalmically to treat allergic and vasomotor rhinitis as well as allergic conjunctivitis. It also stabilizes mast cells, inhibiting the release of inflammatory mediators such as interleukin-6, tryptase, histamine, TNF-alpha, and leukotrienes[1][6]. Epinastine is an H1 receptor antagonist indicated for the prevention of itching associated with allergic conjunctivitis; it also stabilizes mast cells and inhibits both H1 and H2 receptors while preventing the release of pro-inflammatory mediators[3]. Both drugs are small molecules that do not significantly cross the blood-brain barrier. While each drug is approved individually for allergic conditions (primarily rhinitis or conjunctivitis), there are no known marketed products or clinical studies combining these two agents together.
