Azercabtagene zapreleucel is an investigational, allogeneic (off-the-shelf), CD19-directed chimeric antigen receptor (CAR) T cell therapy. Unlike autologous CAR-T therapies that use a patient’s own cells, this product is derived from healthy donor T cells and engineered using gene editing technology to express an anti-CD19 CAR while minimizing the risk of graft-versus-host disease. The therapy specifically targets and kills CD19-expressing tumor cells, which are present in B-cell malignancies such as diffuse large B-cell lymphoma (DLBCL) and other non-Hodgkin lymphomas, as well as in normal B-cells. Azercabtagene zapreleucel is being developed for relapsed/refractory DLBCL, other B-cell non-Hodgkin lymphomas, precursor B-cell lymphoblastic leukemia/lymphoma, mantle cell lymphoma, small lymphocytic lymphoma, Waldenstrom macroglobulinemia, high grade B cell lymphoma, and autoimmune diseases including multiple sclerosis. It utilizes the ARCUS genome editing platform to engineer donor-derived T cells[2][4][5][6][8].
