AZP-3813 is a novel, synthetic 16-amino acid bicyclic peptide that acts as a growth hormone (GH) receptor antagonist. It is designed to bind to the GH receptor and block endogenous GH from activating the receptor, thereby reducing downstream production of insulin-like growth factor 1 (IGF-1) by the liver. This mechanism aims to address acromegaly, a rare endocrine disorder characterized by excessive secretion of GH and elevated IGF-1 levels, often due to pituitary tumors. AZP-3813 is being developed primarily as an add-on therapy for patients with acromegaly who are insufficiently controlled with somatostatin analogues (SSAs). The drug has demonstrated dose-dependent reductions in IGF-1 in preclinical and early clinical studies, with additive effects when combined with SSAs such as octreotide. It features a long circulating half-life (~20–22 hours), supporting its use in chronic disease management[1][2][4][6].
