Overview

**Azvudine** is an orally administered, synthetic fluorinated cytidine nucleoside analogue antiviral developed by Genuine Biotech. Following intracellular phosphorylation to its active triphosphate metabolite, it acts as a nucleoside reverse transcriptase inhibitor against HIV and inhibits viral RNA polymerases, including SARS-CoV-2 RNA-dependent RNA polymerase, by serving as an aberrant nucleotide substrate that disrupts viral nucleic-acid synthesis. Genuine Biotech also characterizes azvudine as a dual-target HIV agent that inhibits HIV-1 reverse transcriptase and viral infectivity factor. Azvudine received conditional approval in China for HIV infection in July 2021 and for adult COVID-19 in July 2022; it is also being clinically investigated as GEN-725 in oncology.
