Overview

Azvudine + PD-1 inhibitor is a combination therapy being developed by Genuine Biotech that pairs the small molecule nucleoside analogue azvudine (also known as GEN-725) with a PD-1 monoclonal antibody. This combination is designed to treat advanced solid tumors, specifically liver cancer and colorectal cancer, by leveraging a synergistic mechanism of action. Azvudine acts as an immunomodulator that targets the DNA-MDSC pathway to reduce the presence of myeloid-derived suppressor cells (MDSCs) within the tumor microenvironment, which in turn promotes the infiltration and activity of T cells and natural killer (NK) cells. The PD-1 inhibitor component blocks the Programmed cell death protein 1 immune checkpoint, reactivating T cells to mount an effective anti-tumor response. Preclinical studies have demonstrated significant efficacy, including 100% complete tumor remission in animal models of liver and colorectal cancer with no recurrence after treatment cessation. In January 2026, the company announced that the clinical trial for this combination in advanced solid tumors was approved by China's National Medical Products Administration (NMPA).
