B-cell activating factor receptor (BAFF-R) chimeric antigen receptor (CAR) T-cell therapy is an autologous cellular immunotherapy designed to target malignant B cells. Developed by the Mayo Clinic, the specific candidate MC10029 utilizes a patient's own T cells genetically engineered to express a CAR targeting BAFF-R, a receptor critical for B-cell survival and consistently expressed on various B-cell malignancies. This approach is particularly relevant for patients with relapsed or refractory disease, including those who have developed resistance to CD19-targeted therapies or exhibit CD19-negative tumor escape. The therapy is currently being evaluated in Phase 1 clinical trials for B-cell hematologic malignancies such as chronic lymphocytic leukemia (CLL).
