**B-cell maturation antigen-targeting chimeric antigen receptor T cells** are a target-specific CAR-T-cell therapy platform rather than one uniquely defined drug product. These therapies use patient-derived or donor-derived T cells genetically engineered to express a chimeric antigen receptor that recognizes B-cell maturation antigen, a plasma-cell surface protein highly expressed in multiple myeloma. BCMA engagement activates the engineered T cells and promotes cytotoxic elimination of BCMA-expressing malignant plasma cells. Individual BCMA CAR-T products differ materially in CAR binding domains, co-stimulatory domains, gene-transfer systems, cell source, manufacturing process, dose, and clinical profile. FDA-approved examples for relapsed or refractory multiple myeloma include idecabtagene vicleucel and ciltacabtagene autoleucel, while the CARLOTTA01 study evaluates the distinct investigational autologous product MDC-CAR-BCMA001 in relapsed or refractory multiple myeloma and diffuse large B-cell lymphoma.
