Overview

B. longum 2012 is a genetically engineered strain of the probiotic bacterium Bifidobacterium longum, developed as a component of an oral protein vaccine platform. It is specifically modified to display the galacto-N-biose/lacto-N-biose I-binding protein (GLBP) on its cell surface, which serves as an anchoring mechanism for tumor-associated antigens. In preclinical research for Wilms' tumor 1 (WT1) immunotherapy, B. longum 2012 is used as a control or intermediate strain to evaluate the efficacy of antigen-expressing versions like B. longum 420. The platform utilizes the bacteria's natural tropism for tumor environments and its ability to stimulate intestinal immunity via Peyer's patches to induce systemic immune responses.
