Overview

B.63521Δ11 gp120 is a recombinant HIV-1 envelope glycoprotein (gp120) immunogen derived from a clade B transmitted founder (T/F) virus and engineered with an 11-amino-acid truncation at the N terminus of the protein. This modification enhances antigenicity of V2 and C1 epitopes—key targets for antibody-dependent cell-mediated cytotoxicity (ADCC)—and prevents dimerization of the protein to optimize manufacturing yield and stability[1][3]. The construct may also include additional mutations in the V3 loop to prevent proteolytic clipping during expression in CHO cells[1]. It is used as a protein boost in experimental HIV vaccine regimens, such as those involving DNA/MVA prime-boost strategies (e.g., GOVX-B11), aiming to elicit robust antibody responses against HIV-1[1][8]. The Δ11 deletion alone has been shown sufficient for enhanced antigenicity and immunogenicity compared to unmodified forms[3].
