B10G5 is a non-blocking monoclonal antibody that selectively binds and neutralizes tumor-derived soluble MHC class I chain–related protein (sMIC), a soluble NKG2D ligand that drives systemic immune suppression in cancer. By sequestering free sMIC without blocking MIC interaction with the NKG2D receptor, B10G5 restores NK cell homeostatic renewal, enhances NK and antigen-specific CD8 T-cell activation and memory formation, stabilizes CD3ζ and TCR signaling in CD8 T cells, and remodels the tumor microenvironment by reducing myeloid-derived suppressor cells and tumor-associated macrophages, thereby inhibiting primary tumor growth and metastatic dissemination in MIC-expressing tumor models.[1][2][3][5] The antibody has been humanized (huB10G5) and evaluated preclinically in MICB-transgenic mouse models and non-human primates, and is being developed primarily for solid tumors that shed sMIC, including prostate cancer, melanoma, and non–small cell lung cancer.[1][2][3][4][5][7]
