B12-8D3 is a bispecific antibody construct designed as a non-binding control for evaluating brain-shuttle technologies in central nervous system research. It is composed of the B12 monoclonal antibody, which targets HIV-1 gp120 and does not bind to endogenous brain antigens, fused to a single-chain Fab (scFab) fragment of the 8D3 antibody. The 8D3 component specifically targets the transferrin receptor protein-1 (TfR1), facilitating transport across the blood-brain barrier (BBB) via receptor-mediated transcytosis. In preclinical studies for Alzheimer's disease, B12-8D3 is often radiolabeled with isotopes such as Zirconium-89 (89Zr) for immuno-PET imaging. It serves as a critical baseline to isolate the effects of BBB shuttling from specific target engagement, allowing researchers to quantify non-specific brain uptake and distribution compared to therapeutic candidates like Adu-8D3.
