B2E5-ADC is an experimental antibody-drug conjugate (ADC) designed to target epithelial-V–like antigen 1 (EVA1), a protein identified as a functional factor specifically expressed in glioblastoma-initiating cells (GICs). The therapeutic candidate consists of the B2E5 monoclonal antibody, which was generated by immunizing mice with an EVA1-Fc fusion protein and selected for its high affinity to human EVA1. In preclinical studies, B2E5-ADC demonstrated potent cytotoxicity against EVA1-expressing cell lines and GICs in vitro. Furthermore, the conjugate showed efficacy in preventing tumorigenesis when administered intracranially in GIC tumor-bearing mouse models. Beyond direct payload delivery, the B2E5 antibody component also mediates cell death through antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). The development of B2E5-ADC is a collaborative effort involving researchers from Hokkaido University and RIKEN.
