Overview

B2M-aTac(dsFv) is a recombinant fusion protein designed for cancer immunotherapy, specifically to overcome tumor immune escape mediated by MHC class I downregulation. The molecule consists of a single-chain MHC class I complex (HLA-A2) genetically fused to a disulfide-stabilized variable fragment (dsFv) of the anti-Tac antibody, which targets the interleukin-2 receptor alpha subunit (CD25). By binding to CD25-expressing tumor cells, B2M-aTac(dsFv) effectively "decorates" the cell surface with HLA-A2-peptide complexes. This artificial presentation of MHC-peptide ligands renders HLA-A2-negative tumor cells susceptible to recognition and lysis by specific CD8+ cytotoxic T lymphocytes (CTLs), such as those targeting the melanoma-associated gp100 peptide. This approach aims to redirect the adaptive immune response toward tumor cells that would otherwise evade detection.
