B440 is an innovative oral cancer vaccine comprised of recombinant *Bifidobacterium* engineered to express the Wilms' tumor 1 (WT1) tumor-associated antigen. Developed by Immunorock, B440 is designed to deliver the WT1 protein to dendritic cells within the gut-associated lymphoid tissue (GALT), thereby inducing a tumor-specific cellular immune response. Preclinical studies demonstrated effective WT1-specific T-cell induction and anti-tumor activity in murine models of urothelial, prostate, and renal cancers. In a Phase 1 clinical trial for metastatic urothelial cancer, B440 exhibited a favorable safety profile and successfully induced WT1-specific immunity, which correlated with improved progression-free survival and enhanced clinical responses upon subsequent pembrolizumab rechallenge.
