Overview

B5V60 is a genetically engineered, thermally responsive recombinant elastin-like polypeptide (ELP) functionalized with five copies of the Bld-1 targeting peptide (SNRDARRC). It is designed as a multivalent drug delivery vehicle specifically targeting the sialyl-Tn (STn) antigen, a tumor-associated carbohydrate antigen overexpressed in bladder cancer and various other malignancies. The ELP backbone, consisting of Val-Pro-Gly-Xaa-Gly repeats, provides biocompatibility and a characteristic inverse temperature phase transition (Tt ~40°C), which allows the polypeptide to remain soluble at physiological temperatures while potentially facilitating localized accumulation or drug release. B5V60 leverages the homology between the Bld-1 peptide and Siglec motifs to achieve high avidity and specificity for STn-expressing tumor cells, significantly outperforming monovalent peptides in cellular uptake and tumor localization.
