Overview

B63521^11 gp120 and IHV01 gp120 are recombinant HIV-1 envelope glycoproteins (gp120) developed for use as protein boosts in heterologous prime-boost HIV vaccine regimens. B63521^11 is a monovalent CD4-induced (CD4i) gp120 protein, while IHV01 is a transmitted/founder (T/F) gp120 protein. In the HVTN 132 clinical trial, these proteins are administered alongside the MVA/HIV62B vaccine boost following a GEO-D02 DNA prime. The goal of including these proteins is to enhance the induction of protective antibody responses, particularly those targeting the V1V2 region of the HIV envelope, which has been associated with reduced risk of infection in previous trials like RV144. The development is led by the National Institute of Allergy and Infectious Diseases (NIAID) in collaboration with the Institute of Human Virology and GeoVax.
