Overview

B7-H3 CAR iNK-T cell is an adoptive cell therapy utilizing invariant natural killer T (iNKT) cells engineered with a Chimeric Antigen Receptor (CAR) targeting B7-H3. This therapy is being investigated for its potential in treating childhood central nervous system (CNS) tumors, such as medulloblastoma. iNKT cells are innate-like T cells that naturally migrate towards CCL2, which is overexpressed in medulloblastoma, potentially overcoming immune cell trafficking limitations into CNS tumors. Additionally, they may neutralize the immune suppressive microenvironment by selectively lysing tumor-associated macrophages. The B7-H3 CAR construct enables these iNKT cells to specifically recognize and target B7-H3 expressing medulloblastoma cells, leading to tumor lysis and improved survival in preclinical models.
