Overview

B7-H3 CAR-T cells are an investigational adoptive cellular immunotherapy consisting of autologous or allogeneic T cells genetically engineered to express a chimeric antigen receptor (CAR) targeting the B7-H3 (CD276) antigen. B7-H3 is a member of the B7 family of immune checkpoint proteins that is frequently overexpressed in various solid tumors, including malignant skin neoplasms, neuroblastoma, and gliomas, while showing limited expression in normal tissues. Upon binding to B7-H3 on the surface of tumor cells, the CAR-T cells undergo activation, proliferation, and secretion of cytotoxic cytokines, leading to the targeted destruction of B7-H3-expressing malignant cells. Early-phase clinical trials, such as those conducted by the Dermatology Hospital of Southern Medical University, are evaluating these cells for the treatment of B7-H3-positive malignant skin tumors and other advanced solid malignancies.
