Overview

B7-H3 CAR-T lambda cells are an experimental chimeric antigen receptor (CAR) T-cell therapy engineered to target B7-H3 (CD276) and simultaneously secrete interferon lambda 2 (IFNλ2). Developed by researchers at the University of North Carolina at Chapel Hill, these cells are designed to treat glioblastoma by overcoming the immunosuppressive tumor microenvironment. Unlike type I interferons, which can cause T-cell apoptosis via autocrine signaling, IFNλ2 signals through the IFNLR1 receptor, which is absent on T cells but present on tumor-associated microglia and certain glioblastoma cells. The secreted IFNλ2 promotes the remodeling of microglia into an antigen-presenting phenotype and reduces the stemness of tumor cells, thereby enhancing the antitumor efficacy and persistence of the CAR-T cells.
