B7-H3 chimeric antigen receptor Epstein-Barr virus-specific T cells (B7H3.CAR-EBVSTs) is an investigational allogeneic, off-the-shelf CAR T-cell therapy designed to treat B7-H3-positive solid tumors. The therapy utilizes Epstein-Barr virus-specific T cells (EBVSTs) as a platform, which naturally possess a narrowed TCR repertoire to minimize the risk of graft-versus-host disease (GvHD) without requiring gene editing. These cells are engineered with a nanobody-based (VHH) chimeric antigen receptor targeting B7-H3 (CD276), a protein overexpressed in various solid malignancies including gastric, breast, and pancreatic cancers. To enhance persistence and efficacy in the immunosuppressive tumor microenvironment, the cells are armored with an engineered serine protease inhibitor, SerpinB9(CAS), and a constitutive IL-7 receptor. This combination aims to protect the cells from allogeneic rejection and activation-induced cell death while promoting proliferation even in the absence of lymphodepletion.
