B7-H3 fusion protein vaccine is a cancer vaccine candidate developed by CK Life Sciences targeting the immune checkpoint ligand B7-H3 (CD276). The vaccine consists of a recombinant fusion protein, produced in E. coli, which combines the extracellular domain of B7-H3 with immune enhancers. It is designed to elicit a humoral immune response, specifically generating neutralizing antibodies that block the interaction of B7-H3 with its receptors, thereby disrupting a key mechanism used by cancer cells to escape immune surveillance. In preclinical studies, such as those presented at AACR 2025, the vaccine demonstrated significant anti-tumor activity in pancreatic cancer models, particularly when used in combination with a PD-L1-targeting vaccine, by promoting the infiltration of CD4+ T cells into the tumor microenvironment.
