Drug intelligence / Profile preview
B7-H3 monoclonal antibodies are a class of targeted immunotherapies designed to bind to B7-H3 (CD276), an immune checkpoint protein belonging to the B7 family. B7-H3 is frequently overexpressed on a wide variety of solid tumors—including prostate cancer, small cell lung cancer, pancreatic cancer, and pediatric brain tumors—as well as on tumor-associated vasculature, while showing limited expression in normal tissues. By binding to B7-H3, these monoclonal antibodies can block B7-H3-mediated immune evasion, inhibit tumor-intrinsic oncogenic signaling, and recruit immune effector cells to mediate antibody-dependent cellular cytotoxicity (ADCC) or antibody-dependent cellular phagocytosis (ADCP). Several B7-H3 monoclonal antibodies, such as enoblituzumab, have been evaluated in clinical trials both as monotherapies and in combination with other checkpoint inhibitors.
