B7-H3-SDIE is an Fc-modified monoclonal antibody designed for the treatment of pancreatic cancer. It targets the antigen B7-H3 (CD276), which is highly expressed in many tumor entities, including pancreatic cancer, with limited expression on healthy tissues. The antibody features amino acid substitutions S239D/I332E in its Fc region, which are engineered to increase its affinity for the Fc receptor CD16 expressed on natural killer (NK) cells. This modification enhances antibody-dependent cellular cytotoxicity (ADCC), leading to effective activation of NK cells against pancreatic cancer cells, resulting in significant tumor cell lysis.
