Drug intelligence / Profile preview
B7-H3-specific CAR T cells are an investigational class of chimeric antigen receptor (CAR) T-cell therapies engineered to target B7-H3 (CD276), an immune checkpoint protein that is highly expressed across various solid tumors—including pediatric brain tumors (such as diffuse intrinsic pontine glioma [DIPG] and diffuse midline glioma [DMG]), neuroblastoma, sarcomas, and adult cancers like glioblastoma and prostate cancer—while showing limited expression in healthy tissues. These therapies involve harvesting autologous or allogeneic T cells, genetically engineering them to express a CAR targeting B7-H3 (often utilizing costimulatory domains like 4-1BB or CD28), and infusing them back into the patient. Several academic institutions and biotechnology companies, such as Seattle Children's Therapeutics, BrainChild Bio, and St. Jude Children's Research Hospital, are actively developing specific B7-H3 CAR-T candidates (e.g., BCB-276, Loc3CAR) administered either systemically or locoregionally (e.g., intracerebroventricularly) to treat refractory solid tumors and central nervous system malignancies.
