B7-H3-targeted microbubbles are a diagnostic ultrasound molecular imaging (UMI) agent consisting of lipid-shelled, perfluorocarbon-filled microbubbles functionalized with ligands, such as antibodies or engineered affibodies, that specifically target B7-H3 (CD276). B7-H3 is a member of the B7 family of immune checkpoint markers and is significantly overexpressed on the vascular endothelial cells of various malignancies compared to normal tissue. Developed primarily by researchers at Stanford University, these microbubbles bind to the tumor neovasculature upon intravenous administration, enabling high-resolution ultrasound imaging for the detection and characterization of cancers. Preclinical studies have demonstrated their utility in identifying breast cancer (including ductal carcinoma in situ and lymph node metastasis), renal cell carcinoma, and enhancing high-intensity focused ultrasound (HIFU) therapy for liver cancer.
