B7-H3-targeting CAR-NK cells are an experimental allogeneic cellular immunotherapy designed for the treatment of various solid tumors. These natural killer (NK) cells are genetically engineered to express a chimeric antigen receptor (CAR) that specifically targets B7-H3 (CD276), an immune checkpoint molecule that is highly overexpressed in many aggressive malignancies, including glioblastoma, neuroblastoma, and non-small cell lung cancer. Developed as an 'off-the-shelf' product, these cells can be derived from healthy donors or cord blood, offering a potential safety advantage over autologous CAR-T therapies by reducing the risk of graft-versus-host disease (GvHD) and cytokine release syndrome. Preclinical studies have demonstrated that these engineered NK cells exhibit potent, antigen-specific cytotoxicity against B7-H3-positive tumor cells, leading to significant antitumor effects and prolonged survival in xenograft models.
