The B7-H3 vaccine is a novel recombinant protein subunit vaccine designed to target B7-H3 (CD276), an immune checkpoint molecule frequently overexpressed in various solid tumors. The vaccine construct features tandem repeats of the FG loop fragment of human B7-H3, a structural motif essential for ligand-receptor interactions, fused to an N-terminal cell-penetrating peptide (CPP) to enhance its immunogenicity. When administered with the Toll-like receptor 9 (TLR9) agonist adjuvant CpG ODN 1826, the vaccine elicits a robust dual immune response, including the production of anti-B7-H3 antibodies and the activation of IFNγ-producing CD4+ and CD8+ T-cells. Preclinical studies in B7-H3-humanized mouse models of colon cancer have demonstrated significant tumor growth inhibition and high rates of complete tumor regression in both prophylactic and therapeutic settings.
