B7H3 CAR iNK cells (specifically developed as FT573 by Fate Therapeutics) are an off-the-shelf, multiplexed-engineered, induced pluripotent stem cell (iPSC)-derived natural killer (iNK) cell therapy designed to target B7-H3 (CD276), a pan-cancer antigen overexpressed in many solid tumors and hematologic malignancies. The therapeutic candidate is engineered with four key functional components: a chimeric antigen receptor (CAR) optimized for NK cell biology that targets B7-H3; a high-affinity, non-cleavable CD16 (hnCD16) Fc receptor to enhance antibody-dependent cellular cytotoxicity (ADCC) when combined with monoclonal antibodies; a membrane-bound IL-15/IL-15 receptor fusion (IL-15RF) to promote cytokine-autonomous persistence and activation; and a CD38 knockout (CD38-KO) to prevent fratricide and improve metabolic fitness in the tumor microenvironment.
