Overview

B7H3 CAR-T refers to a class of chimeric antigen receptor (CAR) T-cell therapies engineered to target the B7-H3 (CD276) antigen, a member of the B7 family of immune checkpoint molecules. B7-H3 is highly overexpressed in various solid tumors—including neuroblastoma, glioblastoma, and gastrointestinal cancers—while maintaining restricted expression in normal healthy tissues, making it a promising target for cellular immunotherapy. These therapies involve collecting a patient's T cells and genetically modifying them to express a CAR that recognizes B7-H3. When re-infused into the patient, these cells bind to B7-H3-expressing tumor cells, triggering T-cell activation and expansion, which leads to the direct lysis of malignant cells. Clinical development is currently focused on overcoming the challenges of the immunosuppressive tumor microenvironment in solid tumors, often exploring combinations with oncolytic viruses or other agents to enhance efficacy.
