B7H3-IL7Ra CAR-T is an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target B7-H3 (CD276), a cell surface protein that is highly expressed in various solid tumors and central nervous system (CNS) malignancies but has restricted expression in normal tissues. To enhance the therapeutic efficacy in the immunosuppressive tumor microenvironment, these CAR-T cells are engineered to co-express a modified version of the Interleukin-7 receptor alpha (IL7Ra), typically a constitutively active version known as C7R. This integrated IL-7 signaling is intended to improve T-cell survival, promote homeostatic expansion, and enhance long-term persistence without the need for exogenous cytokine support. The therapy is currently being evaluated in clinical trials for pediatric and adult patients with recurrent or refractory brain tumors and other B7-H3-positive solid malignancies.
